目的 以传统国标培养法作为参比方法, 评价实时荧光核酸恒温扩增检测(real-time simultaneous amplification and testing, SAT)方法检测食品中金黄色葡萄球菌的能力。方法 依据 GB 4789.10—2016《食品安全国家标准 食品微生物学检验 金黄色葡萄球菌检验》对SAT法检测食品中金黄色葡萄球菌的能力进行评价。分别检测纯培养物、人工污染样品以及实际样品来评价试剂盒的灵敏度、特异性、假阴性率、假阳性率和准确度, 同时采取2种方法的显著性差异分析, 确定最终结果。结果 SAT法灵敏度为100%, 纯培养物检出限为103 CFU/mL, 人工污染检出限灵敏度为103 CFU/mL, 假阴性率为0、假阳性率为2.9%; 金黄色葡萄球菌的特异性符合要求。结论 SAT检测方法灵敏度高、特异性好, 假阳性率、假阴性率、准确度等检测结果全部达标, 并且检测时间短, 适用于食品中金黄色葡萄球菌的快速检测。 |

Objective To evaluate the ability of real-time simultaneous amplification and testing (SAT) method to detect Staphylococcus aureus in food, the traditional national standard culture method is used as the reference method. Methods According to GB 4789.10—2016 National food safety standard-Food microbiological inspection-Staphylococcus aureus, the ability of SAT method to detect Staphylococcus aureus in food was evaluated. The sensitivity, specificity, false-negative rate, false-positive rate and accuracy of the kit were evaluated by detecting pure cultures, artificially contaminated samples and actual samples. At the same time, 2 kinds of methods of significant difference analysis were adopted to determine the final result. Results The sensitivity of SAT method was 100%, and the detection limit of pure culture was 103 CFU/mL. The sensitivity of the detection limit of artificial pollution was 103 CFU/mL. The false-negative rate and false-positive rate were 0 and 2.9%, respectively. The specificity of Staphylococcus aureus met the requirements. Conclusion The SAT detection method has high sensitivity and good specificity. The false positive rate, false negative rate, accuracy and other test results are all up to the standard, and the detection time is short. It is suitable for rapid detection of Staphylococcus aureus in food. |
