目的 对比研究栽培品和野生品太白贝母不同部位核苷类和生物碱类含量。方法 分别提取太白贝母鳞茎、茎、叶中的核苷类和生物碱类成分, 采用高效液相色谱法和紫外-可见分光光度法进行含量测定, 并利用独立样本t检验、聚类分析和相关性分析方法进行评价。结果 太白贝母不同部位中核苷类和生物碱类含量差异性较大, 含量大小整体表现为: 叶>茎>鳞茎; 在9种核苷中腺苷、尿苷、鸟苷含量相对较高, 约占核苷总量的50%~80%; 在鳞茎中胞苷和鸟嘌呤含量相对较低, 约占鳞茎核苷总量的1%~8%; 在茎中尿嘧啶含量最低, 约占茎核苷总量的3%; 而叶中的核苷类成分整体含量较高。总生物碱含量均符合2020年版《中国药典》要求; 独立样本t检验结果表明, 太白贝母野生品与栽培品中核苷类与生物碱类含量无显著性差异(P>0.05), 说明栽培品能代替野生品入药; 聚类分析结果表明, 太白贝母不同部位有效成分含量差异性较大; 相关性分析结果表明, 各有效成分之间存在显著的相关性。结论 太白贝母地上茎叶核苷类和生物碱类含量高于地下鳞茎, 其茎叶存在进一步研究与开发的价值。 |

Objective To compare and study the content of nucleosides and alkaloids in different parts of cultivated and wild Fritillaria taipaiensis. Methods The nucleosides and alkaloids were extracted from bulbs, stems and leaves of Fritillaria taipaiensis, the content was determined by high performance liquid chromatography and ultraviolet-visible spectrophotometry, and evaluated by independent sample t-test, cluster analysis and correlation analysis. Results The content of nucleosides and alkaloids in different parts of Fritillaria taipaiensis was quite different, and the overall content was as follows: leaf > stem > bulb. The content of adenosine, uridine and guanosine in the 9 nucleosides was relatively high, accounting for 50%?80% of the total nucleosides. The content of cytidine and guanine in bulb was relatively low, accounting for 1%?8% of the total nucleosides in bulb. The content of uracil in stem was the lowest, accounting for about 3% of the total nucleosides in stem. The content of nucleosides in leaves was higher. The content of total alkaloids was in accordance with the requirements of Chinese pharmacopoeia (2020 Edition). The independent sample t-test results showed that there was no significant difference in the content of nucleosides and alkaloids between wild and cultivated Fritillaria taipaiensis (P>0.05), indicating that cultivated Fritillaria taipaiensis could replace wild Fritillaria taipaiensis. The results of cluster analysis showed that the content of effective components in different parts of Fritillaria taipaiensis was different. The results of correlation analysis showed that there was a significant correlation between the effective components. Conclusion The content of nucleosides and alkaloids in aboveground stems and leaves of Fritillaria taibaiensis is higher than that in underground bulbs. The stems and leaves of Fritillaria taipaiensis have the value of further research and development. |
