目的 响应面法优化三相分离法提取河蚬汁(Corbicula fluminea juice, CFJ)中活性多糖的工艺。方法 首先对河蚬汁的基本成分进行分析, 然后利用三相分离法(three-phase partitioning, TPP)从河蚬汁中提取多糖, 以硫酸铵质量分数、叔丁醇与河蚬汁体积比、提取温度和时间作为单因素变量, 以多糖提取率和可溶性蛋白含量为指标, 利用单因素实验结合响应面法优化确定TPP提取河蚬汁多糖(CFJPs)的最佳提取条件。结果 优化得到的TPP提取CFJPs的最佳条件为: 硫酸铵质量分数26.0%(m:V)、叔丁醇与河蚬汁体积比1.4:1.0(V:V)、温度37.0 ℃、时间30 min。在最佳提取条件下, CFJPs提取率的模型预测值为49.80%, 实验值为48.05%±1.09%, 实验值和预测值相差1.75%, 表明响应面模型是正确、可行的。结论 本研究可为从河蚬汁中分离提取活性多糖以及实现河蚬汁的综合利用提供参考。 |

Objective To optimize the extraction process of active polysaccharides from Corbicula fluminea juice (CFJ) by three-phase partitioning (TPP) using response surface methodology. Methods The basic components of CFJ were analyzed, and then CFJPs from CFJ were extracted by three-phase partitioning (TPP). The mass fraction of ammonium sulfate, volume ratio of t-butanol to CFJ, extraction temperature and extraction time as single factor variables, polysaccharides yield and soluble protein content as indicators, the single factor experiment and response surface methodology (RSM) were used to determine the optimal extraction conditions for extraction of CFJPs. Results The optimized TPP extraction conditions of CFJPs were as follows: ammonium sulfate mass fraction 26.0% (m:V), volume ratio of t-butanol to CFJ 1.4:1.0 (V:V), temperature 37.0 ℃ and extraction time 30 min. Under the optimal extraction conditions, the predicted value of the extraction yield of polysaccharides was 49.80%, while the experimentally measured value was 48.05%±1.09%. The difference between the experimental value and the predicted value was 1.75%, indicating that the applied RSM model was correct and feasible. Conclusion This study will provide a reference for the separation of active polysaccharides from CFJ and the comprehensive utilization of CFJ. |
