      目的 筛选荞麦及其制品中总黄酮含量测定的可行性方法。方法 以芦丁和槲皮素为标准品, 利用直接法、NaNO2-Al3+-NaOH法、AlCl3法3种比色法扫描标准品及样品的光谱图, 并通过光谱重复性、最大波长下的标准曲线、干扰实验以及精密度、稳定性、加标回收实验对所选方法进行评价。结果 直接法不适合甜荞中总黄酮含量的测定且受酚酸类物质干扰极大, 使得该法检测结果不可靠; NaNO2-Al3+-NaOH法紫外区5个波长的吸光度值重复性较差, 可见区的最大吸收波长易受酚酸类物质的干扰, 该法检测结果偏高; AlCl3法基本不受酚酸类物质的影响, 光谱重复性好、精密度高、稳定性好、平均回收率为90.1%, 在0.0~0.5 mg/mL范围内芦丁的浓度与吸光值呈良好的线性关系, r2为0.999, 显色后2 h内吸光值稳定。结论 AlCl3法可行实用, 适合作为测定荞麦及其制品中总黄酮含量的方法。

      Objective To select a feasible method for the determination of total flavonoids in buckwheat and its products. Methods Rutin and quercetin were used as the standard, 3 kinds of colorimetric methods, namely direct method, NaNO2-Al3+-NaOH method and AlCl3 method, were used to scan the spectrograms of the standard and samples, and the selected method was evaluated through spectral repeatability, standard curve at the maximum wavelength, interference test, precision, stability and standard recovery test. Results The direct method was not suitable for the determination of total flavonoids content in buckwheat and was greatly interfered by phenolic acids, which made the detection result of this method unreliable. The absorbance values of the five wavelengths in the ultraviolet region of NaNO2-Al3+-NaOH method have poor repeatability, and the maximum absorption wavelength in the visible region is easily interfered by phenolic acids. The detection results of this method were relatively high. AlCl3 method is basically unaffected by phenolic acid substances, with good spectral repeatability, high precision, good stability and average recovery of 90.1%. In the range of 0.0 to 0.5 mg/mL, there was a good linear relationship between the concentration of rutin and the absorbance value, r2 was 0.999, and the absorbance value was stable within 2 h after color development. Conclusion AlCl3 method is practical and suitable for determination of total flavonoids in buckwheat and its products.
