目的 运用基质辅助激光解吸电离飞行时间质谱法(matrix assisted laser analytical ionization time of flight mass spectrometry, MALDI-TOF-MS)对9批中药材中相关污染菌进行快速鉴定, 考察其微生物污染情况。方法 参照2015年版《中国药典》四部, 并结合MALDI-TOF-MS对分离出来的菌落进行鉴定。结果 甘草、黄芪、党参的lgTAMC(需氧菌总数, total aerobic microbial count)、lgTYMC(霉菌和酵母菌总数, total yeasts and molds count)分别为4.91~7.04、1.0~4.11, lgTB(耐热菌总数, total thermophilic bacteria count)为1.48~2.65, 而且经过100 ℃、30 min热处理后lgTAMC数值下降百分比均在50%以上。鉴定结果中显示污染概率最大的是成团泛菌、简单芽孢杆菌和地衣芽孢杆菌。结论 甘草、黄芪、党参药材中污染的微生物多为成团泛菌和芽孢杆菌, 在后期药材加工环节, 可以采用有针对性的、有效的处理方法, 从而避免由于原药材本身带来的不利影响。 |

Objective To quickly identify the related contamination bacteria of nine batches of Chinese medicinal materials by matrix assisted laser analytical ionization time of flight mass spectrometry(MALDI-TOF-MS), and investigate its microbial pollution situation. Methods The isolated colonies were identified in reference according to the 2015 edition of the Chinese Pharmacopoeia (Vol. 4) in combination with MALDI-TOF-MS. Results The lgTAMC (total aerobic microbial count) and lgTYMC (total yeasts and molds count) of Glycyrrhizae radix et rhizoma, Astragali Radix and Codonopsis radix were 4.91–7.04 and 1.0–4.11 respectively, while the lgTB (total thermophilic bacteria count) was 1.48–2.65. Besides, the numerical decline percentage of lgTAMC was above 50% after heat treatment at 100℃ for 30 min. The identification results showed that the most probable contamination was Pantoea agglomerans, Bacillus simplex and Bacillus licheniformis. Conclusions The contaminated microorganisms in the medicinal materials of Glycyrrhizae radix et rhizoma, Astragali Radix and Codonopsis radix are mostly Pantoea agglomerans and Bacillus, and targeted and effective treatment methods can be adopted in the later medicinal material processing links to avoid adverse effects caused by the original medicinal materials. |
