目的 研究抗菌肽(I1WL5W)对创伤弧菌细胞膜的抑制作用。方法 通过测定最低抑菌浓度(minimum inhibitory concentration, MIC)、圆二色光谱(circular dichroism, CD)实验、细胞膜去极化实验、SYTOX绿色摄取实验, 检测抗菌肽对创伤弧菌的抗菌活性、对细胞膜的破坏能力及作用机制。结果 抗菌肽(I1WL5W)对创伤弧菌的MIC值为2.08 μmol/L, 其能够与细菌膜结合, 具有膜去极化活性, 具有抗菌活性, 对弧菌细胞膜有较强的破坏能力。结论 抗菌肽通过破坏创伤弧菌膜完整性的方式, 诱导细菌细胞死亡。 |

Objective To study the inhibitory effect of antimicrobial peptide (I1WL5W) on Vibrio vulnificus cell membrane. Methods Minimum inhibitory concentration (MIC), circular dichroism (CD) test, cell membrane depolarization test, SYTOX green uptake test, and flow cytometry test for bacterial membrane rupture were used to determine the antibacterial activity of antimicrobial peptides against Vibrio vulnificus, their ability to destroy cell membranes, and their mechanism of action. Results The MIC value of antibacterial peptide (I1WL5W) against Vibrio vulnificus was 2.08 μmol/L, which could bind to the bacterial membrane, had membrane depolarizing activity and antibacterial activity, and had strong ability to destroy the membrane of Vibrio. Conclusion Antimicrobial peptides can induce bacterial cell death by destroying the membrane integrity of Vibrio vulnificus. |
