目的 建立高效液相色谱串联质谱法检测黄花鱼中硫黄素T的方法。方法 样品以甲醇作为萃取溶剂, 经C18反相色谱柱分离, 以甲醇和0.1%甲酸酸化的0.02 mol/L乙酸铵作为流动相梯度洗脱, 流速为 0.30 mL/min, 采用电喷雾离子源, 正离子多反应检测扫描方式监测, 基质匹配标准曲线法定量。结果 硫黄素T在1~100 ng/mL范围内具有良好的线性, 方法定量限为1.2 μg/kg, 回收率为85.5%~93.8%, 相对标准偏差为0.5%~3.1%。结论 该检测方法操作简单, 结果准确, 可应用于黄花鱼中硫黄素T的日常检测。 |

Objective To establish a method for the determination of thioflavin T in yellow croaker by high performance liquid chromatography-tandem mass spectrometry. Methods Samples were extracted with methanol as solvent, separated by C18 reverse phase chromatography with gradient elution using methanol and 0.1% formic acidified 0.02 mol/L ammonium acetate as the mobile phases. The flow rate was 0.30 mL/min. The electrospray ionization source was used. The positive ion multiple reaction detection scanning method was used for monitoring. The matrix matching standard curve method was used for quantitative analysis. Results The thioflavin T had good linearity in the range of 1?100 ng/mL. The limit of quantitation was 1.2 μg/kg, the recoveries were 85.5%?93.8%, and the relative standard deviation were 0.5%?3.1%. Conclusion The method is simple and the result is accurate. It can be applied to the daily detection of thioflavin T in yellow croaker. |
