      目的 研究灵芝多糖(Ganoderma lucidum polysaccharides, GLP)的最优提取工艺, 并研究其抗氧化活性。方法 通过单因素和响应面法优化提取工艺参数, 获得GLP; 将GLP进行乙醇分级得到GLP30、GLP60和GLP80 3个组分; 采用还原力、DPPH自由基清除试验和羟自由基清除试验评估GLP及3个组分的抗氧化活性。结果 灵芝多糖最优提取条件为: 温度86 °C, 液料比50:1 (mL/g), 时间142 min, 实际提取得率为1.71%; 抗氧化活性试验结果表明: GLP及GLP30、GLP60和GLP80均具有一定的抗氧化活性, 均呈现浓度-活性依赖性, 其中GLP80还原力最强, GLP清除DPPH自由基和羟自由基的能力最强。结论 响应面法有效优化了GLP的提取, 灵芝多糖具有较好的抗氧化活性, 值得进一步研究及开发利用。

      Objective To study the optimal extraction process of Ganoderma lucidum polysaccharides (GLP), and its antioxidant activity. Methods The extraction process of GLP was optimized by single factor and response surface methodology. GLP30, GLP60, GLP80 were fractionated from GLP by alcohol fractional precipitation technique, and their antioxidant activities were evaluated on the basis of the reducing power, DDPH radicals scavenging activity and hydroxyl radicals scavenging activity. Results The optimal extraction conditions for GLP were as follows: extraction temperature 86 ℃, liquid-to-material ratio 50:1 (mL/g), extraction time 142 min. Under this condition, the actual extraction yield was 1.71%. The results of antioxidation experiment showed that the 4 polysaccharides had certain antioxidant activities and showed concentration dependent effect. GLP80 had the strongest reducing power, and the DPPH radicals and hydroxyl radicals scavenging rates of GLP were the highest. Conclusion Response surface method can effectively optimize the extraction of GLP. GLP and GLP80 show better antioxidant activity, which deserves further research and development.
