      目的 探究甲壳类水产品过敏原的荧光免疫层析检测方法中面临的基质影响问题。方法 以原肌球蛋白作为检测目标, 选取3种水产品加工场景中的常见基质: 鱼肉、肉制品(肠)、酱类作为研究对象, 从量子点荧光探针、免疫识别反应和层析性能3个方面分析基质干扰的影响途径, 提出改进措施。结果 基质条件对T线荧光强度有消减, 尤其在鱼肉和肠基质下对检测结果的判读影响较大; 荧光探针在基质条件下信号稳定。结论 基质对于检测的影响主要来自于对抗原抗体的结合反应产生的干扰。本研究进一步探讨了基质效应的研究思路, 为食物过敏原快速检测形式在实际应用情景下的发展提供理论支持。

      Objective Toinvestigate the matrix effects of rapid detection methods based on fluorescent lateral-flow immunoassayfor crustacean aquatic products allergens. Methods Using tropomyosin as the detection target, 3kinds of food matrix in common aquatic product processing scenarios were selected as research objects in this experiment, which were fish, meat products (sausage), and sauces.Influence pathways and improvement measures were analyzed from aspects of quantum dots-based fluorescent probes, immunorecognition reactions and chromatographic properties, respectively. Results The interference of matrix made a decrease on the fluorescence intensity of T-line, especially under the fish and sausage matrix. Fluorescent probes had stable signals under matrix conditions. Conclusion The influence of the matrix on the detection mainly comes from the interference caused by the binding reaction of the antigen and antibody. This article further explores the idea of investigating matrix effects and provides theoretical support for the development of rapid food allergen detection forms in practical applications.
