      目的 通过响应面法优化高压脉冲电场辅助提取蓝靛果花青素的工艺参数。方法 以长白山野生蓝靛果为原料, 乙醇作为溶剂, 使用紫外分光光度计全波长扫描获取花青素最大吸收波长, 以此为依据测定花青素含量, 探究高压脉冲电场辅助提取法提取花青素时电场强度、脉冲数、乙醇体积、料液比4个因素对花青素提取量的影响, 采用响应面法优化其提取工艺, 并与传统试剂浸提法最优参数、提取量进行对比。结果 高压脉冲电场辅助提取花青素的最佳工艺参数为: 乙醇体积分数67%, 电场强度20 kV/cm, 脉冲数10个, 液料比1:78 g/mL, 此时蓝靛果花青素提取量为34.20 mg/g。结论 高压脉冲电场辅助提取花青素经优化后具有溶剂消耗少、提取时间短、花青素提取量多的优势, 有良好的发展前景。

      Objective To optimize the process parameters of Lonicera edulis anthocyanin extraction assisted by high voltage pulsed electric field(PEF) by response surface method. Methods Taking wild Lonicera edulis in Changbai mountain as raw material, ethanol as solvent, the maximum absorption wavelength of anthocyanins was obtained by full wavelength scanning of UV spectrophotometer. Based on this, the content of anthocyanins was determined. The effects of electric field intensity, pulse number, ethanol volume and solid-liquid ratio on the extraction of anthocyanins by high voltage pulsed electric field assisted extraction method were studied. The results were optimized by response surface method. The optimum extraction parameters and extraction amount were compared with traditional reagent extraction method. Results The optimum extraction conditions of anthocyanins from Lonicera edulis were as follows: ethanol concentration 67%, electric field intensity 20 kV/cm, pulse number 10, and liquid-solid ratio 1:78 g/mL, and the extraction amount of anthocyanins from Lonicera edulis under these conditions was 34.20 mg/g. Conclusion The optimized high voltage pulsed electric field assisted extraction of anthocyanins has the advantages of low solvent consumption, short extraction time, and large amount of anthocyanins, and has good development prospects.
