      目的 建立榅桲果实和种子中委陵菜酸、熊果酸和齐墩果酸等三萜酸薄层色谱分离鉴定方法。方法 以甲醇为溶剂, 超声提取榅桲果实和种子, 对展开系统、点样量、薄层板、显色剂等薄层色谱条件进行考察, 确定最佳薄层鉴定条件。结果 以点样量为4 μL, 将点于高效硅胶GF254板上的样品点浸入1%碘二氯甲烷溶液中预处理, 以甲苯: 乙酸乙酯: 冰醋酸(体积比: 14:4:0.5)为展开剂, 10%硫酸乙醇溶液来显色, 在366 nm下观察, 委陵菜酸、熊果酸和齐墩果酸被同时检识, 分离效果好。结论 薄层色谱法分辨率高、操作简便、结果可靠, 可用于榅桲属中三萜酸类化合物的分离和鉴别。
英文摘要:
      Objective To establish thin layer chromatography method to separate and identificate tormentic acid, ursolic acid and oleanolic acid in Cydonia oblonga fruit and seeds. Methods With methanol as solvent, the quince fruit and seeds were extracted by ultrasound. The optimum thin layer chromatography conditions were determined by investigating the development system, the amount of samples, the thin layer plate and the chromogenic agent. Results The 4 μL sample point on the high efficiency silica gel GF254 plate was immersed in 1% iodine-dichloromethane solution for pretreatment, toluene: ethyl acetate: glacial acetic acid (volume ratio, 14:4:0.5) was used as the developing agent, and developed the color with 10% ethanol solution of sulfuric acid. At 366 nm, the tormentic acid, ursolic acid and oleanolic acid were detected at the same time, and effectively separated on the thin layer chromatography plate. Conclusion The method has the advantages of high resolution, simple operation and reliable results. It can be used for the separation and identification of triterpenoid acids in the Cydonia oblonga.
