目的 构建基于天麻高效液相色谱(high performance liquid chromatography, HPLC)指纹图谱法结合化学计量学分析对硫磺熏蒸的天麻进行识别。方法 参照《中华人民共和国药典》一部(2015年版), 采用高效液相色谱-梯度洗脱法测定硫磺熏蒸天麻的指纹图谱, 通过中药色谱指纹图谱相似度评价系统软件(2012版)进行指纹图谱信息统计, 将信息输入SIMCA-P14.0软件进行主成分分析法和偏最小二成判别分析。结果 该方法能较好的分离天麻的活性成分, 依据检测结果确定了18个共有指纹峰, 由于未硫熏组和硫熏组天麻样品的指纹图谱存在差异性, 利用化学计量学方法可以正确识别硫熏天麻。结论 利用高效液相指纹图谱结合化学计量分析可准确识别硫磺熏蒸的天麻。 |

Objective To establish high performance liquid chromatography (HPLC) fingerprint combined with chemometrics method for analysis of Gastrodia elata Bl before and after sulfur-fumigated for its discrimination. Methods According to the Pharmacopoeia of the People’s Republic of China (2015 version), HPLC method coupled with a gradient eluted program was used to analyze the fingerprint of Gastrodia elata Bl samples fumigated with sulfur. The operating standard of similarity evaluation system for chromatographic fingerprint of Chinese materia medica (version 2012) was used to compare the quality of different batches of GR samples. The principal component analysis (PCA) and the partial least squares (PLS) was applied to analyze the chromatograms by SIMCA-P14.0. Results The chemical constituents of Gastrodia elata Bl were optimally separated, and 18 fingerprint peaks in common were confirmed. There was apparent difference between the fingerprints of the samples before and after sulfuring. Chemo metrics can be used for the research on the component changes caused by sulfur-fumigation of Gastrodia elata Bl. Conclusion The developed HPLC fingerprints combined with chemo metrics can accurately identify sulfur-fumigation of Gastrodia elata Bl. |
