      目的 研究显齿蛇葡萄总黄酮含量测定的最佳提取条件。方法 以显齿蛇葡萄为原料, 用乙醇-水为溶剂, 加热超声浸提, 并通过试验优化出最佳的提取工艺条件, 以芦丁为标准品, 选择波长415 nm, 采用三氯化铝分光光度法进行总黄酮含量的测定。结果 在提取温度60 ℃、提取时间30 min、乙醇浓度95%、提取次数3次时, 黄酮的提取效率最高; 在此方法下, 标液浓度在0.5~56 μg/mL范围内, 浓度与吸光值呈良好线性关系, 回归方程为Y=0.0287X, r2=0.9998, 方法最低检出限为2 mg/kg, 样品加标的平均回收率为98.30%, 标准偏差(relative standard deviation, RSD)分别为0.94%、1.62%; 通过实验室间结果比对, 检测结果稳定, RSD均<2.0%。结论 该方法精密度及稳定性好, 可以较好地提取显齿蛇葡萄中的总黄酮。

      Objective To study on the best extraction conditions of total flavonoids in ampelopsis grossedentata. Methods Ampelopsis grossedentata was used as raw material, total flavonoids were extracted by heated ultrasound with ethanol-water as solvent. The best extraction conditions were optimized through experiments. Total flavonoids were determined by AlCl3 spectrometric method at 415 nm wave-length with Rutin as standard. Results The extraction efficiency of flavonoids was the highest when the extraction temperature was 60 ℃, the extraction time was 30 min with 95% ethanol concentration under 3 times of extraction. The regression equation was Y=0.0287X with a good linearity (r2=0.9998) between the absorbance and concentration in the range of the standard sample concentration from 0.5 to 56 μg/mL. The detection limit was 2 mg/kg. The average recovery rate was 98.30%, and RSD (relative standard deviation) was 0.94% and 1.62%. There were stable results by comparing interlaboratorys, and the RSD was less than 2%. Conclusion The method has good precision and stability, and can better extract the total flavonoids from ampelopsis grossedentata.
