目的 评价VITEK MS对副溶血性弧菌(Vibrio parahaemolyticus, VP)的识别能力。方法 培养平板选用弧菌显色平板和血平板, 处理方法选用甲酸辅助裂解法(方法1)和直接涂板法(方法2)。每个菌株的不同培养平板与处理方法的组合分别用RUO软件和IVD软件进行鉴定, 鉴定结果用配对卡方检验分析培养平板和处理方法对鉴定结果的影响。用RUO软件对图谱做相似性分析。结果 IVD软件可以对139株VP的不同培养平板和方法组合100%鉴定到种的水平, RUO软件对方法1和方法2处理的属水平鉴定率分别不超过60%和45%, 种水平的鉴定率分别不超过17%和9%, 2种处理方法结果差异具有统计学意义(P<0.05)。同一种处理方法2种平板的结果差异无统计学意义(P>0.05)。VP图谱之间的相似性为45%?90%, 其和溶藻弧菌图谱之间的相似性为54%?78%。结论 IVD软件对VP有较好的鉴定效果, RUO软件对其鉴定效果不理想, 甲酸辅助裂解法可以稍微提高鉴定率。VP图谱和溶藻弧菌图谱有交叉聚类的现象。 |

Objective To evaluate the ability of VITEK MS to recognize Vibrio parahaemolyticus(VP). Methods The culture plates selected the Vibrio chromogenic plate and blood plate, and the treatment methods selected the clinical auxiliary resolution method (Method 1) and the direct plating method (Method 2). The combination of different culture plates and treatment methods of each strain was identified by RUO software and IVD software respectively. The identification results were analyzed by paired chi-square test to analyze the influence of culture plates and treatment methods on the identification results. The similarity analysis of spectra was performed on RUO software. Results Totally 139 VP strains were 100% successfully identified to the species level in different combinations of plates and methods by IVD software. The identification rate of genus level treated by method 1 and method 2 did not exceed 60% and 45% respectively, and to species level less than 17% and 9% respectively by RUO software. The difference between the 2 treatment methods was statistically significant (P<0.05). There was no significant difference in the results of the same treatment between the two kinds of plates (P>0.05). Intraspecific similarity among VP spectra was 45%?90%, and the interspecific similarity between VP and Vibrio alginolyticus (VAL) was 54%?78%. Conclusions The IVD software has a good identification effect on VP, but RUO software is not ideal for its identification. The method of formic acid assisted lysis could slightly increase the identification rate. There is a cross clustering phenomenon between VP and VAL spectra. |
