目的 建立高效液相色谱法(high performance liquid chromatography, HPLC)测定破布木果水提物中迷迭香酸的含量。方法 用高效液相色谱法, 色谱条件: Kromasil C18柱(4.6 mm150 mm, 5 μm), 流动相为甲醇-1.5%乙酸水(38: 62), 流速为1.0 mL/min, 柱温25 ℃, 检测波长为330 nm, 检测破布木果水提物中迷迭香酸的含量。结果 迷迭香酸峰面积与进样量在15.625~1000 μg/mL范围内呈良好的线性关系(r=0.9996), 平均回收率为98.68%, 相对标准偏差为2.13%(n=6)。结论 HPLC法具有操作简便、结果准确可靠的优势, 可用于检测破布木果水提物中迷迭香酸含量。 |

Objective To establish a method for determination of rosmarinic acid content in water extract of Cordia dichotoma fruits by high performance liquid chromatography (HPLC). Methods HPLC was used to determine the content of rosmarinic acid in water extract of Cordia dichotoma fruits. Chromatographic conditions were as follow: chromatographic column was Kromasil C18 column (4.6 mm150 mm, 5 μm), mobile phase was methanol-1.5% acetic acid water (38:62), flow rate was 1.0 mL/min, column temperature was 25 ℃, detection wavelength was at 330 nm. Results The peak area of rosmarinic acid and the injection volume showed a good linear relationship (r=0.9996) in the range of 15.6251000 μg/mL, the average recovery was 98.68%, and the relative standard deviation was 2.13% (n=6). Conclusion This method is simple, accurate and reliable, and it is feasible for the determination of rosmarinic acid content in water extract of Cordia dichotoma fruits. |
