      目的 优化石榴皮中不溶性膳食纤维酶法提取的最优条件。方法 以果胶酶和木瓜蛋白酶水解石榴皮为原料, 以石榴皮不溶性膳食纤维得率为指标, 对液料比、酶添加量、酶解温度以及酶解时间4个单因素对石榴皮不溶性膳食纤维得率影响的基础上进行L9(34)的正交优化试验。结果 在液料比为20:1(m/V)的条件下, 果胶酶添加量0.9%, 酶解温度55 ℃, 酶解时间65 min; 木瓜蛋白酶添加量0.6%, 酶解温度50 ℃, 酶解时间45 min, 在此条件下, 石榴皮不溶性膳食纤维的得率可达31.87%±0.27%。结论 酶法提取石榴皮不溶性膳食纤维得率高, 条件温和、安全性高、利于环保。

      Objective To optimized the process conditions for extracting pomegranate skin insoluble dietary fiber by enzymatic method Methods Pectinase and papain were used to hydrolyze pomegranate peel, and the yield of pomegranate peel insoluble dietary fiber was used as the experimental index. The orthogonal optimization experiment of L9(34) was carried out on the basis of the influence of liquid-to-liquid ratio, enzyme addition amount, enzymatic hydrolysis temperature and enzymatic hydrolysis time on the yield of pomegranate skin insoluble dietary fiber. Results Under the condition of liquid-solid ratio (m/V) of 20:1, the pectinase addition amount was 0.9%, the enzymatic hydrolysis temperature was 55 ℃, and the enzymatic hydrolysis time was 65 min; the papain addition amount was 0.6%, the enzymatic hydrolysis temperature was 50 ℃, and the hydrolysis time was 45 min. Under these conditions, the yield of pomegranate skin insoluble dietary fiber can reach 31.87%±0.27%. Conclusion Enzymatic extraction of pomegranate skin insoluble dietary fiber has high yield, mild conditions, high safety and environmental protection.
