目的 比对2种荧光观察方法检测生活饮用水中大肠埃希氏菌的适用性、灵敏度及稳定性。方法 使用全自动菌落计数器与传统多管发酵法2种方法对大肠埃希氏菌EC-MUG肉汤管进行荧光观察测试、分析。结果 仪器法较传统法灵敏, EC-MUG肉汤管培养4 h后使用仪器即可观察到蓝色荧光, 6 h后呈现深蓝色荧光, 而传统法在6 h时荧光现象微弱呈淡蓝色, 10 h后荧光现象逐渐明显呈现蓝色。分别使用传统法与仪器法观察10组水样结果无差异。结论 观察水中大肠埃希氏菌荧光现象时可使用仪器法和传统法, 但仪器法更有优势, 灵敏度高, 操作简单, 值得推广。 |

Objective To compare the applicability, sensitivity and stability of the 2 kinds of fluorescence observation methods for the detection of Escherichia coli in drinking water. Methods Automatic instrumental method and traditional multi-tube fermentation method were used to observe and analyze the fluorescence of Escherichia coli EC-MUG broth tube. Results The instrumental method was more sensitive than traditional method. The instrumental method showed that blue fluorescence could be observed after 4 h, while it was more obvious after 6 h. Traditional method showed that blue fluorescence gradually became light blue after 6 h, while it was gradually obvious after 10 h. There was no difference between traditional method and instrumental method in stability test. Conclusion Instrumental method and traditional method can be used to observe the fluorescence of Escherichia coli in water, but the instrumental method has more advantages, higher sensitivity and simpler operation, which is worth popularizing. |
