      目的 利用响应面分析法优化刺参中金刚烷胺的检测。方法 在单因素基础上, 确定采用1%乙酸乙腈作为提取溶剂, 以1%乙酸乙腈的添加量、超声时间、PSA吸附剂的添加量、C18吸附剂的添加量为影响因素, 以金刚烷胺的回收率为响应值, 根据Box-Behnken试验设计原理对刺参中金刚烷胺提取条件进行响应面分析, 优化前处理条件。结果 1%乙酸乙腈为15 mL, 超声提取10 min, PSA吸附剂为250 mg时刺参中金刚烷胺的回收率最大。在此优化条件下进行验证性试验, 测得金刚烷胺的回收率为118%, 与预测值相对误差为9.62%。实际检测刺参样品20个, 2个样品检出, 分别为4.3、1.6 μg/kg。结论 该优化前处理方法具有良好的可行性。

      Objective To optimize the detection of amantadine from Apostichopus japonicus by response surface methodology. Methods Based on single factor test, 1% acetonitrile acetate was used as extraction solvent, the adding amount of 1% acetonitrile acetate, ultrasonic time, PSA adsorbent and C18 adsorbent were taken as the influencing factors, and the yield of amantadine was taken as the response value. Box-Behnken experiment design combined with response surface analysis were used to optimize the detection of amantadine from Apostichopus japonicus. Results The optimal extraction process conditions were as follows: 1% acetonitrile acetate was 15 mL, ultrasonic time was 10 min and PSA adsorbent was 250 mg for the recovery of amantadine from Apostichopus japonicus. Under this conditions, the recovery of amantadine was 118% with a 9.62% error compared with the actual value. The 20 samples were actually tested, and 2 samples were detected of amantadine, which were 4.3 and 1.6 μg/kg. Conclusion It shows that the optimized pretreatment method is feasible.
