目的 探讨阿里红多糖(Fomes officinals Ames polysaccharide, FOPS)及多糖纯化组分(FOP-a)对小胶质细胞(BV-2)炎症反应的抑制作用。方法 采用β淀粉样蛋白毒性片段(amyloid-β25-35, Aβ25-35)(40 μg/mL)诱导小胶质细胞BV-2活化, 建立炎症反应模型, 利用四甲基偶氮唑蓝法检测细胞存活率, 并用酶联免疫法考察FOPS及FOP-a对Aβ25-35诱导的BV-2细胞产生的IL-1及TNF-α炎症因子的影响; LDH试剂盒检测BV-2细胞中乳酸脱氢酶活性表达变化; 采用QPCR方法检测FOPS及FOP-a对BV-2细胞中IL-6、TNF-a、MCP-1、iNOS、COX-2及NF-κB等基因mRNA表达的影响; 采用蛋白印迹方法检测FOPS及FOP-a对BV-2细胞中iNOS及COX-2蛋白表达的影响。结果 FOPS及FOP-a可抑制Aβ25-35诱导的BV-2细胞IL-1及TNF-α炎症因子的释放和LDH活性表达(P<0.05)。FOPS及FOP-a显著抑制IL-6、TNF-a、MCP-1、iNOS、COX-2及NF-κB等基因mRNA表达(P<0.05)。FOPS及FOP-a抑制促炎因子iNOS、COX-2的蛋白表达(P<0.05)。结论 阿里红多糖及纯化组分可明显减轻Aβ25-35诱导下产生的小胶质细胞BV-2的神经炎症反应。 |

Objective To investigate the inhibitory effects of Fomes officinals Ames polysaccharides (FOPS) and Fomes officinals Ames polysaccharides components (FOP-a) on the inflammatory response in BV-2 microglial cells induced by Aβ25-35. Methods The mouse BV-2 microglial cells were cultured and activated by Aβ25-35, and the inflammatory response model was established. The cell survival rate was measured by tetramethyl azolium blue method. The effects of FOPS and FOP-a on IL-1 and TNF-α inflammatory factors produced by BV-2 cells induced by a Aβ25-35 were investigated by enzyme-linked immunoassay. The effects on the release capacity of LDH were determined by LDH kit. The effect of FOPS, FOP-a on the mRNA expression of IL-6, TNF-a, MCP-1, iNOS, COX-2, NF-ΚB in BV-2 microglial cells were determined by QPCR. The effects of FOPS and FOP-a on iNOS and COX-2 protein expression in BV-2 microglial cells were detected by Western blot. Results FOPS and FOP-a could inhibited the release of IL-1 and TNF-α inflammatory cytokines and the active expression of LDH in BV-2 microglial cells induced by Aβ25-35 (P<0.05). FOPS and FOP-a inhibited the mRNA expression of IL-6, TNF-a, MCP-1, iNOS, COX-2, NF-ΚB significantly (P<0.05). FOPS and FOP-a inhibited the protein expression of iNOS and COX-2 (P<0.05). Conclusion Polysaccharides components FOPS and FOP-a from Fomes officinals Ames can inhibit the neuroinflammatory response in BV-2 microglia cells. |
