      目的 建立高效液相色谱法测定牛奶磷脂酰胆碱(phosphatidylcholine, PC)含量的分析方法。方法 对高效液相色谱法测定条件进行优化, 包括流动相选取、柱温选取和流速选取问题。其中, 在V(乙腈):V(甲醇):V(水)=600:200:5的条件下进行不同浓度PC标准溶液的响应值分析试验; 在60 ℃柱温条件下进行PC分离效果试验; 选取流速为0.6 mL/min, 进行PC溶液洗脱分析试验。最后采用高效液相色谱法, 进行牛奶磷脂酰胆碱含量测定, 并根据试验结果绘制标准曲线。结果 PC的平均加标回收率在90.8%~96.53%之间, 相对标准偏差是2.82%~9.81%。5份牛奶样品测定含量的相对误差在5%以内, 满足要求。结论 该方法准确、可行、高效, 适用于牛奶磷脂酰胆碱含量的测定。

      Objective To establish a method for the determination of phosphatidylcholine (PC) in milk by high performance liquid chromatography. Methods Firstly, the experimental conditions of high performance liquid chromatography were optimized, including the selection of mobile phase, column temperature and flow rate. Under the conditions of V (acetonitrile):V(methanol):V(water)=600:200:5, the response value analysis test of PC standard solution with different concentrations was carried out; the separation effect test of PC was carried out under the condition of column temperature of 60 ℃; and the elution analysis test of PC solution was carried out at the flow rate of 0.6 mL/min. Finally, the content of phosphatidylcholine in milk was determined by high performance liquid chromatography, and the standard curve was drawn according to the test results. Results The average recoveries of PC were between 90.8% and 96.53%. The relative standard deviations (RSDs) of PC were 2.82%?9.81%. The relative errors of the determination of PC content in 5 kinds of milk samples were less than 5%, which met the requirements. Conclusion This method is accurate, feasible and effective, which is suitable for the determination of PC in milk.
