目的 比较预混合饲料中维生素A的3种提取方法。方法 抽取10个预混合饲料样品, 比较皂化提取法、直接提取法、二甲基亚砜(dimethyl sulfoxide, DMSO)提取法实验过程和维生素A的检测结果。结果 皂化提取法提取过程时间(t=130.4 min)较长, 步骤(21步)繁杂, 较其他2种提取方法检测结果准确性较差, 平行性较差。直接提取法与DMSO提取法提取过程时间较短, 步骤较简便, 检测结果准确性较好, 平行性较好。结论 直接提取法可以作为快速检测预混合饲料中维生素A含量的手段, 但检测结果使用受到GB/T 17817-2010《饲料中维生素A的测定高效液相色谱法》限制。皂化提取法可用于低含量复杂样品检测。DMSO法提取法, 相较于皂化提取法, 操作简便, 测试快速, 检测值准确性较好, 平行性较好, 能过有效弥补直接提取法与皂化提取法的缺点。 |

Objective To compare the 3 kinds of extraction methods of vitamin A in the premixed feed. Methods Ten samples of premixed feed were extracted. The feasibility of the experimental process and extraction efficiency of vitamin A of saponification extraction method, direct extraction method and DMSO extraction method were compared. Results The process of saponification extraction was time-consuming and complicated, the detection result was less accuracy and the parallelism was worse than the other two methods. The process of direct extraction method and DMSO extraction method less time were needed, the steps was simple, the detection result was more accurate, and the parallelism was better. Conclusion Direct extraction can be used as a rapid method for the determination of vitamin A in premixed feed. However, the determination of vitamin A in feed was limited by GB/T 17817-2010 Determination of vitamin A in feed high efficiency liquid chromatography. Saponification extraction method can be used for the detection of complex samples with low content. Compared with saponification extraction method, DMSO extraction method is easy to operate, quick to test, accurate detection value and good parallelism, which can make up the shortcomings of direct extraction method and saponification extraction method. |
