目的 建立胶体金免疫层析法快速检测水产品中孔雀石绿总量的残留。方法 用柠檬酸三钠还原法制备胶体金并标记孔雀石绿单克隆抗体。基于竞争抑制反应原理, 在优化条件下建立胶体金免疫层析法。 结果 水产品中孔雀石绿总量的检测限为2 μg/mL, 与结构类似物结晶紫的交叉反应不明显。实际样本检测结果与液相色谱-串联质谱法检测结果的差异较小。结论 该方法快速、有效, 可满足现场检测水产品中隐性孔雀石绿及孔雀石绿的残留。 |

Objective To develop a method for the rapid determination of total malachite green (MG) residues in aquatic products by colloidal gold immunochromatography assay (GICA). Methods Colloidal gold was prepared by sodium citrate reduction method, which was marked malachite green monoclonal antibody. Under optimized conditions, a sensitive GICA was developed based on competitive binding immunoassay. Results The limit of detection was 2 μg/mL for total amount of leucomalachite green (LMG) residues in aquatic products, and the cross-reaction with the structural analog crystal violet was not obvious. The difference between the actual sample test results and the liquid chromatography-mass spectrometry results was small. Conclusion This method is rapid and efficient, which can meet the need of on-site rapid detection of LMG and MG residues in aquatic products. |
