目的 建立高效液相色谱-蒸发光法测定一种保健品中盐酸氨基葡萄糖含量的分析方法。方法 采用Waters Xbridge Amide柱(4.6 mm×150 mm, 3.5 μm), 以乙腈:0.2%三乙胺水(V:V, 75:25)为流动相, 流速为 1.0 mL/min, 柱温为40 ℃。蒸发光散射检测器检测条件为漂移管温度105 ℃, 载气N2, 压力12.5 bar。结果 盐酸氨基葡萄糖进样量在0.12886~1.03088 μg时与峰面积呈良好线性关系(r2=0.9998), 平均加样回收率为101.91%, 相对标准偏差为1.43%。结论 此方法简单、准确, 精密度和重现性良好, 可用于保健品中盐酸氨基葡萄糖含量的测定。 |

Objective To establish a method for the determination of glucosamine hydrochloride in health food by high performance liquid chromatography evaporative light-scattering detector (HPLC-ELSD). Methods The HPLC-ELSD was performed on Waters Xbridge Amide (4.6 mm×150 mm, 3.5 μm). With acetonitrile-0.2% Triethylamine (V:V, 75:25) as mobile phase, the flow rate was 1.0 mL/min, and the column temperature was 40 ℃. The test conditions of evaporative light scattering detector (ELSD) were as follow: drift tube temperature 105 ℃, N2 as carrier gas and gas pressure 12.5 bar. Results There was a good linear relationship between the peak area and the sample volume of glucosamine hydrochloride at 0.12886~1.03088 g (r2=0.9998), the average recovery rate was 101.91%, and the relative standard deviation (RSD) was 1.43%. Conclusion This method is simple, accurate and repeatable, which is suitable for the determination of the content of glucosamine hydrochloride in health food. |
