      大黄鱼(Larinichthys crocea)主要分布在中国沿海和近海,是一种具有很高经济价值的鱼类。但在亲鱼繁殖和网箱水产养殖中易受许多病原体与疾病的侵害。目的:本文旨在探究苦味肽对大黄鱼先天免疫系统的影响,以期利用酶水解苦味肽提高鱼类在水产养殖中的免疫能力。方法:分别利用木瓜蛋白酶、胰蛋白酶、胃蛋白酶,碱性蛋白酶和风味蛋白酶酶解鮸鱼肉制备苦味最高的苦味肽,通过感官评定最终确定胃蛋白酶水解产生的苦味肽苦味最高。以幼大黄鱼为实验对象,分别设置4个不同的组,对其采取腹腔注射苦味肽,并分别观察8周后进行采血检测,以白细胞/总血细胞的平均比率和溶菌酶活性为指标评价苦味肽对大黄鱼先天免疫系统的作用效果。结果表明:白细胞/总血细胞的平均比率:对照组为14.6%,低剂量组(0.6mg/鱼)为29.3%,中剂量组(1.2mg/鱼)为35%,高剂量组(2.4mg/鱼)为30%。在白细胞对乳胶颗粒的体外吞噬作用实验中,与检测分析白细胞数量的顺序一致,对照组与测试组的吞噬指数分别为1.15(0mg/L),1.88(0.4mg/L),3.84(0.8mg/L),5.94(1.2mg/L)。测定溶菌酶在60min内的相对裂解活性分别为0.17,0.101,0.307和0.198。结论:以上结果表明,与对照组相比苦味肽可以提高大黄鱼的免疫力,当注射剂量为1.2mg时,吞噬与溶菌酶活性都显著升高,可初步确定最佳注射剂量为1.2mg。综上,可推断苦味肽具有刺激炎症,通过多途径有效地提高天然免疫的作用,可以降低大黄鱼疾病的发生,值得进一步研究。

      The large yellow croackers (Larinichthys crocea) are mainly present in the Chinese coast and near seas with high economic importance, but vulnerable to many diseases, especially in the breeding and aquaculture. Objective: To investigate the effects of bitter peptides on the innate immune system of Pseudosciaena crocea, in order to improve the immunity of fish in aquaculture by enzymatic hydrolysis of bitter peptides. Methods :Total 120 Juvenile of large yellow croackers were divided into 4 different groups in 200/300 liter of seawater tank, respectively, and were observed for 3 months before and after different treatments. The bitter peptides from pepsin hydrolysis was applied because its highest bitter sensory scores. The blood of fish from the different groups was collected and tested for different immune parameters to evaluate the effectiveness of bitter peptides as immune stimulants after administration for 8 weeks. Results: The average ratio of leukocytes/total blood cells for control was found at 14.6%, for the low dose of 0.6 mg/fish was at 29.3%, and for middle dose of 1.2 mg/fish was at 35%, and high dose of 2.4 mg/fish was at 30%. Phagocytic index was1.15 for control, 1.88 for 0.4mg/ml, 3.84 for 0.8mg/ml, and 5.94 for 1.2mg/ml, respectively. The lysozyme assay shows the relative lysis activity at 60 minutes were 0.17, 0.101, 0.307 and 0.198, respectively in the same order as in phagocyte assay. Conclusion: The results showed that bitter peptides can be used as immune boosters for the yellow croakers and the best dose was 1.2 mg/fish as both leukocytes and lysozyme activity increased significantly compared to the control group. According to the results obtained we suggest that the incorporation of middle dose (1.2 mg/fish) of the bitter peptides into fish feeds may reduce the fish diseases in aquaculture.
