目的 建立高效液相色谱法测定食品中胭脂红酸含量的分析方法。方法 用2 mol/L盐酸提取样品中的胭脂红酸, 经混合型阴离子交换固相萃取柱净化后, 以0.02 mol/L磷酸二氢铵和甲醇作为流动相, 采用二极管阵列检测器进行检测。结果 胭脂红酸在0.2~100 μg/mL的范围内, 浓度与峰面积的线性关系良好, 相关系数(r2)为0.9999; 方法检出限为0.3 mg/kg, 方法定量限为1.0 mg/kg, 在10、50、200 mg/kg 3个添加水平时, 平均加标回收率为84.8%~97.3%(n=6), 相对标准偏差为2.7%~7.9%。结论 该方法操作简单, 具有较高的灵敏度和准确性, 适用于食品中胭脂红酸的测定。 |

Objective To establish a method for the determination of carmine acid in food by high performance liquid chromatography (HPLC). Methods The sample was extracted by 2 mol/L hydrochloric acid, and purified by mixed anion exchange solid phase extraction column. The mobile phase were 0.02 mol/L ammonium dihydrogen phosphate and methanol, and the samples were detected by diode array detector. Results Carminic acid had good linear relationship in the range of 0.2-100 μg/mL, and the correlation coefficient (r) was 0.9999. The limit of detection and limit of quantitation were 0.3 mg/kg and 1.0 mg/kg, respectively. The recoveries at 3 spiked levels (10, 50, 200 mg/kg) were 84.8%-97.3%, with the relative standard deviations (RSDs) were 2.7%-7.9%. Conclusion This method is simple to operate, and has the advantage of high sensitivity and accuracy, which is suitable for the determination of carmine red acid in food. |
