      目的 探究香菇醇提物的最佳提取工艺, 比较醇提物不同极性部位的抗氧化活性。方法 在单因素和正交实验结果的基础上, 利用Box-Benhnken法进行3因素3水平中心组合设计, 使用响应面法优化香菇醇提物提取的工艺条件。分别采用石油醚、氯仿、乙酸乙酯和乙醇对醇提物进行萃取, 以清除1,1-二苯基-2-三硝基苯肼自由基(DPPH?)、羟自由基(?OH)的能力及总还原力作为香菇多糖抗氧化能力的评估标准, 筛选出最佳抗氧化活性部位。结果 本研究建立了数学模型, 得到二次多项回归方程, 并获得最佳醇提物提取工艺条件: 料液比1:9 (g/mL), 时间2.23 h, 温度40 ℃, 酒精浓度51.50%。提取得率实际值为16.79%, 预测值为17.54%, 误差为4.27%。经数据分析拟合良好, 模型显著。抗氧化活性结果表明, 4种不同极性部位的醇提物均有一定的抗氧化活性, 其中乙酸乙酯相活性最高。结论 该法能有效优化香菇醇提物的提取, 乙酸乙酯相 的抗氧化活性最高, 值得深入开发利用。

      Objective To investigate the optimal extraction process of the ethanol extract of Lentinus edode, and compare the antioxidant activity of the different polar parts of the alcohol extracts. Methods On the basis of single factor and orthogonal test results, the Box-Benhnken method was used to design a 3-factor and 3-level center combination, and the response surface method was used to optimize the extraction conditions of the Lentinus edodes extract. The ethanol extracts were leached with petroleum ether, chloroform, ethyl acetate and ethanol, respectively. The ability to remove 1,1-diphenyl-2-trinitrophenylhydrazine free radicals (DPPH?), hydroxyl radicals (?OH) and total reducing power were used as the evaluation criteria for the antioxidant capacity of Lentinus edodes polysaccharides, and the good antioxidant active sites were selected. Results A mathematical model was established to obtain a quadratic multiple regression equation, and the optimal extraction conditions of the alcohol extract were obtained: The ratio of material to liquid was 1:9 (g/mL), the time was 2.23 h, the temperature was 40 ℃, and the alcohol concentration was 51.50%. The actual yield value was 16.79%, the predicted value was 17.54%, and the error was 4.27%. The data analysis fitted well and the model was significant. The results of antioxidant activity showed that all the alcohol extracts of the 4 different polar sites had certain antioxidant activities, among which the ethyl acetate phase had the highest activity. Conclusion This method can effectively optimize the extraction of the ethanol extract of the Lentinus edode, and the ethyl acetate phase has the highest antioxidant activity, which is worthy of further development and utilization.
