目的 研究原花青素(proanthocyanidins, PC)对反式脂肪酸(trans fatty acids, TFA)致雄性小鼠生殖损伤的保护作用。方法 以昆明成年雄性小鼠为实验对象, 60只小鼠随机分为5组: 正常对照组、TAF组、低、中、高剂量PC保护组, 实验结束后, 测定相关指标。结果 PC可以使小鼠生精细胞数量减少, 使其活率、活力升高, 从而降低畸形率; 经PC保护的染毒的小鼠睾丸乳酸脱氢酶同工酶(lactate dehydrogenase, LDH-X)、γ-谷氨酰转肽酶(glutamyl transpeptidase, γ-GGT)的活性均增加, 表明PC能拮抗TFA致睾丸酶的损害作用; 小鼠睾丸组织的丙二醛(malonic dialdehyde, MDA)含量在给予TFA染毒后明显增高, 说明TFA能对小鼠睾丸组织造成损伤作用, 而经PC保护后, 能降低TFA引起睾丸损伤小鼠MDA含量, 表明PC可明显抑制TFA所致自由基导致的睾丸脂质过氧化损伤作用, 提高机体清除氧自由基能力; 超氧化物歧化酶(superoxide dismutase, SOD)、γ-谷氨酰转肽酶(glutamyl transpeptidase, γ-GGT)活力降低, 而经PC保护后逐渐升高, 反映脂质过氧化产物在睾丸组织内大量堆积从而造成的生殖损害。结论 TFA染毒可以导致雄性小鼠生殖系统损伤。PC干预能够改善TFA致雄性小鼠生殖损伤的作用。 |

Objective To study the protective effects of proanthocyanidins (PC) on reproductive damage induced by trans fatty acids (TFA) in male mice. Methods With Kunming adult male mice as experimental subjects, 60 mice were randomly divided into 5 groups: normal control group, trans fatty acid exposure group, low, medium and high dose PC protection group. The relevant indicators were determined after the experiment. Results PC could reduce the number of spermatogenic cells in mice, increase their viability and vitality, and reduce the rate of deformity. The lactate dehydrogenase (LDH-X) and glutamyl transpeptidase (γ-GGT) activity of PC-protected mice testicular increased, which indicating that PC could antagonize the damage of TFA-induced testicular enzyme. The malonic dialdehyde (MDA) content in mouse testis tissue significantly increased after TFA administration, indicating that TFA could cause damage to the testicular tissue of mice. |
