      目的 建立超高效液相色谱-串联质谱法(ultra performance liquid chromatography-tandem mass spectrometry, UPLC-MS/MS)测定蕨菜中原蕨苷的含量。方法 以DNA加合物为指导分离原蕨苷对照品, 采用Agilent Zorbax Extend C18色谱柱(2.1 mm×100 mm, 1.8 μm), 以乙腈和超纯水为流动相, 梯度洗脱, 流速为 0.4 mL/min, 柱温30 ℃, 多反应模式监测, 内标法定量。结果 原蕨苷在进样质量浓度范围内呈现良好的线性关系(r2=0.9967), 平均加标回收率为91.17%~93.03%, 相对标准偏差为5.60%~6.91%。结论 该方法快速、灵敏度高、专属性好, 可用于测定蕨菜中原蕨苷的含量。

      Objective To establish a method for determination of ptaquiloside in Pteridium aquilinum by ultra performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS). Methods The DNA adduct was used as a guide to separate the ptaquiloside reference substance. The samples were separated on a Agilent Zorbax Extend C18 column (2.1 mm×100 mm, 1.8 μm) at the flow rate of 0.4 mL/min by gradient elution with acetonitrile and ultrapure water as mobile phase. The temperature of the column was maintained at 30 ℃. The detection was carried out by multiple reaction monitoring and quantitatively analyzed by internal standard method. Results Ptaquiloside showed good linear relationships within the investigated concentration ranges (r2=0.9967), the average recoveries were 91.17%-93.03%, and the relative standard deviations (RSDs) were 5.60%-6.91%. Conclusion This method is rapid, sensitive and specific, which can be used to determine the content of ptaquiloside in Pteridium aquilinum.
