目的 研究已建立的脱氧雪腐镰刀菌烯醇时间分辨荧光免疫定量检测体系(包括脱氧雪腐镰刀菌烯醇时间分辨荧光免疫层析检测卡和荧光定量检测仪)的适用性。方法 检测20个阴性样本中脱氧雪腐镰刀菌烯醇含量, 确定该检测体系的检出限和定量限; 对6组含不同浓度脱氧雪腐镰刀菌烯醇的小麦阳性样本和6组含不同浓度脱氧雪腐镰刀菌烯醇的玉米阳性样本进行检测, 确定方法的准确性和台间差; 重复6次检测含中等浓度脱氧雪腐镰刀菌烯醇的阳性样本和连续12 h检测国家检测限浓度的阳性样本, 确定系统的重复性和稳定性。结果 该体系的检出限为154 μg/kg, 定量限为414 μg/kg, 并且检测结果与液质联用方法定值结果无显著性差异, 仪器台间差无显著差异; 测值的精密度未超过国家标准规定要求。结论 本研究验证的时间分辩荧光定量检测体系能准确检测玉米和小麦样本中脱氧雪腐镰刀菌烯醇含量。 |

Objective To investigate the applicability of the established quantitative detection system for the time-resolved fluoro-immunoassay (TRFIA) of deoxynivalenol, including TRFIA quantitative test strips and quantitative detector. Methods The limit of detection and limit of quantitative of the detection system were determined by detecting the content of deoxynivalenol in 20 negative samples. Totally 6 groups of wheat positive samples with different concentrations of deoxynivalenol and 6 groups of maize positive samples with different concentrations of deoxynivalenol were detected, in order to determine the accuracy and inter-station difference of the method. Through 6 times of detection of positive samples with middle concentration of deoxynivalenol, and detection of the positive samples with the national detection limit concentration for 12 consecutive hours, the repeatability of the system was determined. Results The limit of detection for this system was 154 μg/kg and limit of quantitation was 414 μg/kg, and there were no significant differences between detection value and fixed value and between instruments. The precision of the measured value did not exceed the requirements of national standards. Conclusion This verified deoxynivalenol time-resolved fluoro-immunoassay detection system can accurately detect the content of deoxygenated fusarium oxygenate in corn and wheat samples. |
