      目的 建立绞股蓝提取物皂苷成分的高效液相色谱测定法, 并测定市场上绞股蓝提取物皂苷含量。方法 比较甲醇、乙醇、水做提取溶剂及超声、回流提取方式下的皂苷得率。色谱柱为Waters Synnetry C18 (4.6 mm×250 mm, 5 μm); 柱温: 35 ℃; 流速 1.0 mL/min; 检测波长为203 nm。结果 以甲醇为溶剂, 超声 30 min皂苷含量较高。绞股蓝皂苷XLⅨ、绞股蓝皂苷XLⅥ、绞股蓝皂苷A、皂苷XⅦ分别在2.605~ 130.24 μg/mL(r2=0.9992)、2.205~110.24 μg/ mL(r2=0.9995)、1.981~99.04 μg/ mL(r2=0.9992)、1.794~89.68 μg/mL (r2=0.9995)范围内线性关系良好, 平均回收率分别为98.5%~100.4%、97.3%~101.4%、98.0%~101.1%及99.1%~100.6%。结论 本方法灵敏、准确、专属性强, 能准确有效地检测绞股蓝提取物中4种皂苷的含量。

      Objective To establish a method for determination of saponins in Gynostemma pentaphyllum extract by high performance liquid chromatography (HPLC). Methods The extraction rate of saponin was compared with methanol, ethanol and water as extraction solvents and ultrasonic and reflux extraction methods. The chromatographic gradient elution procedure acetonitrile-water was carried out on a Waters Synnetry C18 column (250 mm×4.6 mm, 5 μm). The detection wavelength was set at 203 nm, flow rate was 1.0 mL/min, and the column temperature was 35 ℃. Results The saponin yield was the highest when the methanol was used as the extract solvent and ultrasound was used as the extract method. The linear ranges of gypenoside XLⅨ, XLⅥ, A and XⅦ were within 2.605-130.24 μg/mL(r2=0.9992), 2.205-110.24 μg/mL(r2=0.9995), 1.981-99.04 μg/mL(r2=0.9992), and 1.794-89.68 μg/mL(r2=0.9995), respectively. The average recoveries were 98.5%-100.4%, 97.3%-101.4%, 98.0%-101.1%, and 99.1%-100.6%, respectively. Conclusion The method is sensitive, accurate and specific, which can be used for the determination of 4 kinds of saponins in Gynostemma pentaphyllum extract.
