目的 建立高效液相色谱法(high performance liquid chromatography, HPLC)测定包头地区蒙古黄芪中毛蕊异黄酮葡萄糖苷含量的方法, 对比包头市固阳县、土默特右旗和达茂旗蒙古黄芪中毛蕊异黄酮葡萄糖苷的含量差别, 为更好评价本地区蒙古黄芪品质提供依据。方法 色谱柱为Tnature C18色谱柱(4.6 mm× 250 mm, 5 μm), 以乙腈-0.2%甲酸溶液为流动相梯度洗脱, 流速1.0 mL/min, 柱温30 ℃, 检测波长254 nm。 结果 在10.23~102.3 μg/mL范围内, 毛蕊异黄酮葡萄糖苷与峰面积呈良好线性关系, r2=0.9994, 平均加标回收率为96.93%~98.73%, 相对标准偏差(relative standard deviation, RSD)为1.56%~2.27%。结论 该方法简单、快速、可重复性好, 可用于测定蒙古黄芪中毛蕊异黄酮葡萄糖苷的含量, 且3个旗县的蒙古黄芪中毛蕊异黄酮葡萄糖苷的含量有差别, 其中以土默特右旗所产蒙古黄芪中其含量最高, 且生长年限不同的蒙古黄芪中毛蕊异黄酮葡萄糖苷含量也有差别。本研究为今后内蒙古地区蒙古黄芪的栽培和应用提供科学、可靠的依据。 |

Objective To establish a method for the determination of isoflavones glucoside in Radix Astagali by high performance liquid chromatography (HPLC) and compare the content of isoflavone glucoside in Radix Astagali of Guyang, Tumote Youqi and Damao, so as to provide basis for better evaluation of Radix Astagali quality in Baotou. Methods The column was TnatureC18 column (4.6 mm×250 mm, 5 μm) and acetonitrile-0.2% formic acid was as the mobile phase. The flow rate was 1.0 mL/min, the column temperature was 30 ℃ and the detection wavelength was 254 nm. Results In the range of 10.23-102.3 μg/mL, there was a good linear relationship between the content and the peak area of isoflavone glucoside, with r2 of 0.9994, the average recoveries were 96.93%-98.73% and the relative standard deviation (RSD) were 1.56%-2.27%. Conclusion The method is fast, simple and reproducible, which can be used for the determination of isoflavones glucoside in Radix Astagali. The content of isoflavone glucoside in Radix Astagali is different in the 3 producing areas and the highest content of isoflavone glucoside produced by Tumote Youqi and there are differences in the content of isoflavone glucoside in different growth years. Thus it will provide a scientific and reliable basis for the cultivation and application of Radix Astagali in Inner Mongolia. |
