目的 研究新食品原料宝乐果(Borojo)防晒(sun protection factor, SPF)以及抗紫外线(ultraviolet, UV))照射修复损伤的作用功效。方法 按实验目的将豚鼠和SPF级(specific pathogen free, SPF)小鼠设置空白对照组、基质组、宝乐果组、阳性对照组、宝乐果+阳性对照共5组。按实验内容配方制成各组实验用护肤乳液。仪器法预测试各组SPF值, 动物体内实验在各组豚鼠背部按2 mg/cm2的量分别涂抹护肤乳液, 以功率1346 μW/cm2、波长300~400 nm的紫外灯照射紫外光。测量各组最小红斑量(minimal erythemal dose, MED), 并根据SPF值判定其实际防晒等级。各组小鼠背部皮肤分别均匀涂抹护肤乳液后以波长300~400 nm的长波紫外线(ultraviolet radiation A, UVA)模拟日光紫外照射致损伤; 观察皮肤外观损伤表现; 用试剂盒法测定皮肤组织中超氧化物歧化酶(super oxide dismutase, SOD)和丙二醛(malondialdehyde, MDA)的含量; 统计分析各组数据结果。结果 仪器测试标准SPF15, 对照14.7±2.1, 宝乐果护肤乳液17.7±1.6, 宝乐果+阳性对照组21.0±1.0。体内实验SPF值宝乐果组14.1±1.0, 阳性对照组13.8±1.8, 宝乐果+阳性对照组17.2±2.0。动物皮肤外观同基质组相比, 涂抹照射7 d后, 宝乐果组有轻度红斑少量角质化皱纹, 但未脱皮。宝乐果+阳性对照组红斑较浅无角质化。涂抹照射14 d后, 宝乐果组和宝乐果+阳性组皮肤角质化和红斑渐渐消失同空白对照组。皮肤匀浆测试同基质组比, 宝乐果组小鼠皮肤匀浆中SOD含量明显增加(P<0.05), 宝乐果+阳性对照组显著增加(P<0.01)。宝乐果组小鼠皮肤匀浆中MDA含量明显降低(P<0.05), 宝乐果+阳性对照组显著降低(P<0.01)。 结论 在体内体外试验中, 新食品原料宝乐果配制的护肤乳液具有优良的防晒效果和明显地增强晒后损伤修复作用。 |

Objective To investigate the sun protection factor (SPF) of Borojo fruit and its protective activity in repairing ultraviolet A (UVA)-induced radiation damage. Methods According to the aims of the study, guinea pigs and specific pathogen free (SPF) mice were randomly divided into 5 groups, which were blank control group, matrix component group, Borojo group, positive control group and Borojo plus positive material control group. The different skin lotions used in these 5 groups were prepared by individual recipes. The SPF values of each group were tested by the following strategy. The skin lotions were applied to back of guinea pigs of different groups at 2 mg/cm2. The skin was irradiated with ultraviolet light at a power of 1346 μW/cm2 and a wavelength of 300-400 nm. The minimal erythemal dose (MED) was measured and sunscreen level was determined by the SPF values. The skin lotions were applied to the back of mouse from each group before UVA-induced damage by radiation in the wavelength range of 300–400 nm. The damage of skin appearance caused by radiation was observed. The level of SOD and MDA in the skin tissues were determined by the detection kits. The data were analyzed statistically. Results After measurement, the data indicated that SPF values of standard group, control group, Borojo skin lotion group, and Borojo plus positive group were 15, 14.7±2.1, 17.7±1.6, and 21.0±1.0, respectively. In vivo experiment, SPF values of Borojo group, positive control, Borojo plus positive material group were 14.1±1.0, 13.8±1.8, and 17.2±2.0, respectively. After 7 d of radiation and treatment, mouse skin of Borojo group displayed mild erythema with a little of keratinized wrinkles and without peeling when compared with that of matrix component group. In Borojo plus positive material group, the erythema was mild and no keratinization was observed. After 14 d of UV exposure and treatment, the skin keratinization and erythema were gradually disappeared in Borojo and Borojo plus positive material group, the appearance of back skin of these two groups were almost the same as the blank group. Compared with matrix component group, the levels of SOD and MDA in skin homogenate were significantly higher than those in Borojo group (P<0.05), and SOD value was dramatically increased in positive material control group (P<0.01). Conclusion According to the in vivo and in vitro experiments, the skin care emulsion which is formulated with novel food ingredient Borojo fruit, exhibit extraordinary protective effects in sun protection and repairing skin damage caused by UV radiation. |
