目的 建立液相色谱-串联质谱法(liquid chromatography-tandem mass spectrometry, LC-MS/MS)测定巴西绿蜂胶制品中咖啡因含量的方法。方法 样品经50%甲醇溶液超声提取、Agilent C18色谱柱分离, 以乙腈-0.1%甲酸溶液为流动相进行等度洗脱, 采用液相色谱-串联质谱在正离子模式下检测, 外标法定量。结果 本方法能较好地分离样品中的咖啡因, 在0.8~4.0 μg/mL浓度范围内呈现良好的线性关系, 平均回收率为98.61%~104.72%, 相对标准偏差(relative standard deviation, RSD)为2.3%。结论 本方法准确性好、灵敏度高、重现性好, 适用于巴西绿蜂胶制品中咖啡因含量的测定。 |

Objective To establish a method for determination of caffeine content in Brazilian green bee propolis products by liquid chromatography-tandem mass spectrometry (LC-MS/MS). Methods Samples were ultrasonically extracted with 50% methanol solution, separated by Agilent C18 column with acetonitrile-0.1% methanoic acid solution as the mobile phase for gradient elution, detected by LC-MS/MS in the positive ion mode and quantified by external standard method. Results Caffeine could be separated well, it showed good linear relationship in the range of 0.8~4.0 μg/mL, the average recoveries were 98.61%~104.72%, and the relative standard deviation (RSD) was 2.3%. Conclusion The established method has good accuracy, high sensitivity and good reproducibility, which is suitable for the determination of caffeine content in Brazilian green bee propolis products. |
