目的 鉴定湛江等鞭金藻中主要类胡萝卜素的种类。方法 采用有机溶剂提取湛江等鞭金藻中类胡萝卜素, 利用硅胶柱层析与薄层层析等方法对湛江等鞭金藻中主要类胡萝卜素进行了分离纯化。利用薄层色谱、可见吸收光谱与高效液相色谱-质谱联用等检测手段对湛江等鞭金藻中主要类胡萝卜素组分y1、y2 和y3进行了分析鉴定。结果 结合薄层色谱与定性测试的结果并与标准品对照, 确定湛江等鞭金藻类胡萝卜素组分分y1 和y2 别为β-胡萝卜素和岩藻黄素。结合可见吸收光谱、化学定性测试和液质联用结果推断组分y3为岩藻黄醇。结论 湛江等鞭金藻所含的主要类胡萝卜素分别是β-胡萝卜素、岩藻黄素和岩藻黄醇。 |

Objective To identify the species of the main carotenoids from Isochrysis zhanjiangensis.Methods Carotenoids from Isochrysis zhanjiangensis were extracted by organic solvent. Carotenoids were separated and purified by column and thin-layer chromatography. The major carotenoids fractions y1, y2 and y3 from
Isochrysis zhanjiangensis were analyzed and identified by thin-layer chromatography (TLC), visible spectrum and high performance liquid chromatography-mass spectrometry (HPLC-MS). Results Fractions y1 and y2 were identified as β-carotene and fucoxanthin, based on the results of TLC, chemical tests and comparing results with standard carotenoids. Combined with the results of visible absorption spectrum, chemical test and LC-MS, the component of y3 was deduced to be fucoxanthinol. Conclusion The major carotenoids from Isochrysis zhanjiangensis are β-carotene, fucoxanthin and fucoxanthinol, respectively. |
