目的 探讨卧式螺旋连续动态逆流提取容舒口服液浸膏的优化工艺条件。方法 建立高效液相色谱(high performance liquid chromatography, HPLC)测定容舒口服液浸膏提取液中阿魏酸含量和斯皮诺素含量的方法, 以阿魏酸、斯皮诺素含有量及浸膏得率的综合评分为评价指标, 采用单因素实验、L9(34)正交设计实验, 对粉碎度、溶剂倍数、提取时间、提取温度4 个因素进行考察, 确定最优水提取工艺。结果 HPLC法测定容舒口服液浸膏提取液中阿魏酸含量和斯皮诺素含量的条件为色谱柱: Ultimate XB-C18(GD)(4.6 mm×250 mm, 5 μm), 流动相: 乙腈-0.1%冰醋酸(V:V, 12:88), 流速: 1.0 mL/min, 柱温: 30 ℃, 检测波长330 nm。较优的水提取工艺为: 粉碎度35 目, 溶剂倍数10 倍, 提取时间90 min, 提取温度65 ℃。容舒口服液浸膏提取液中阿魏酸和斯皮诺素含量分别为0.5580 mg/g 和0.5978 mg/g, 浸膏得率为26.89%。结论 优选的容舒口服液浸膏水提取工艺可行, 连续动态逆流提取技术可用于容舒口服液浸膏的提取, 并且有利于工业化生产。 |

Objective To explore the optimum technological conditions for extracting the extract of Rongshu oral liquid by the horizontal spiral dynamic continuous countercurrent extraction. Methods The high performance liquid chromatography (HPLC) method was established for determination of ferulic acid content and spinosin content
of Rongshu oral liquid extract. Taking the comprehensive score of spinosin, ferulic acid content and extract yield as the evaluation indexes, the single factor experiment and L9(34) orthogonal design experiment were conducted to investigate 4 factors of comminution degree, solvent multiple, extraction time and extraction temperature to
determine the optimal water extraction process. Results The conditions of HPLC method to determine the content of ferulic acid and spironine in the extract of the liquid extract were as follows: the chromatographic column was ultimate XB-C18 (GD) (4.6 mm x 250 mm, 5 μm), the mobile phase was acetonitrile-0.1% glacial acetic acid (V:V,
12:88), the flow rate was 1 mL/min, the column temperature was 30 ℃, and the detection wavelength was 330 nm.The optimum water extraction process was as follows: the comminution degree was 35 mesh, the multiple of solvent was 10 times, the extraction time was 90 min, and the extraction temperature was 65 ℃. The content of ferulic acid and spigno in Rongshu oral liquid extract solution were 0.5580 mg/g and 0.5978 mg/g, and the extraction rate was 26.8867%. Conclusion The optimized extraction process of Rongshu oral liquid is feasible. Continuous dynamic countercurrent extraction process can be applied to extract the extract of Rongshu oral liquid, and is conducive to industrial production. |
