      目的 建立103株28种血清型沙门氏菌16SrRNA的进化树, 了解沙门氏菌进化关系, 为基于16SrRNA序列分析对沙门氏菌鉴定分型提供理论基础。方法 使用细菌基因组DNA提取试剂盒提取细菌DNA, 聚合酶链反应(polymerase chain reaction, PCR)扩增16SrRNA基因片段后进行测序, 将序列提交到Genebank和RDP核糖体数据库进行比对, 下载相关参考序列, 使用MegAlign软件Clustal-V法构建进化树。结果 根据16SrRNA同源性能够把103株沙门氏菌聚类成不同群, 部分菌株可以鉴定到血清型, 部分菌株 可以在血清型内进一步区分, 虽然与传统血清学分型不能一一对应, 但是能形成独立的分型系统。 结论 16SrRNA基因序列分析是对沙门氏菌进行鉴定分型及溯源分析的有效方法。

      Objective To establish the phylogenetic tree of 103 strains of 28 serotype Salmonella 16SrRNA and understand the evolution of Salmonella, in order to provide a theoretical basis for the analysis of Salmonella identification based on 16SrRNA sequence analysis. Methods Bacterial DNA was extracted using bacterial genomic DNA extraction kit. The DNA sequencing was performed after the polymerase chain reaction (PCR). The sequences were submitted to the Genebank and RDP ribosomal database for comparison, and the related reference sequences were downloaded and the phylogenetic tree was constructed using the MegAlign software Clustal-V method. Results According to the homology of 16SrRNA, 103 strains of Salmonella were clustered into different groups, some strains could be identified as serotype, and some strains could be further distinguished within serotype, although they were not corresponding one by one with the traditional serological typing, but they could form independent typing system. Conclusion Sequence analysis of 16SrRNA is an effective method for the identification and traceability of Salmonella.
