      目的 建立洁净室环境微生物16SrRNA序列数据库为洁净室环境, 评估实验室质量控制, 为检出菌溯源分析提供依据。方法 采集洁净环境中微生物, 使用MegAlign软件Clustal-V法、MEGA6软件NJ法构建进化树, 分析其种属特异性。结果 本研究共采集到25株微生物, 16SrRNA序列分析结果判定葡萄球属8株占32%, 芽孢杆菌属12株占48%, 5株菌分别属于其他5个属。表面微生物有7株, 占28%, 沉降菌8株, 占32%, 浮游菌10株, 占40%。结论 基本了解洁净室环境微生物构成, 初步建立洁净室环境微生物16SrRNA序列数据库, 为今后实验室质量控制和检出菌溯源提供基础数据和方法。
英文摘要:
      Objective To establish a 16SrRNA database of environmental microbiology in clean room, and to provide basis for environmental evaluation, laboratory quality control and traceability analysis of pathogens. Methods The microorganisms in clean room were collected, and the phylogenetic tree was constructed by using MegAlign software Clustal-V methods and MEGA6 software NJ methods, and their species specificity was analyzed. Results A total of 25 strains of microorganisms were collected in this study. The results of 16SrRNA sequence analysis showed that 8 strains were Staphylococcus accounting for 32%, 12 strains were Bacillus accounting for 48%, and the others belonged to other 5 genus. There were 7 strains of surface microorganisms, accounting for 28%, 8 strains of settling bacteria, accounting for 32%, and 10 strains of airborne microbe, accounting for 40%. Conclusion Basic knowledge of clean room environmental microorganism composition was made, and 16SrRNA database of environmental microbiology in clean room was preliminarily established, which can provide basic data and method for laboratory quality control and detection of bacteria traceability.
