      目的 建立酶比色法快速测定禽蛋中胆固醇含量的分析方法。方法 以酶比色法与分光光度计联用, 在波长505 nm处直接测定经皂化、提取、浓缩和酶工作液催化后的禽蛋样品中胆固醇含量。结果 本试验考察了酶比色反应的最优显色温度(37 ℃)和显色时间(15 min), 并在最优试验条件下测得线性范围为0~2.0 mg/mL, 曲线回归方程Y=0.3207X+0.0891, 相关系数R为0.9992, 检出限为1.0 mg/100 g, 回收率达到90.43%~98.12%, 相对标准偏差小于2.45%。结论 本方法操作简单, 具有很好的精密度与准确度, 适合鸡蛋、鸭蛋、鹅蛋、鸽蛋等全蛋样品或蛋黄样品中胆固醇含量的快速检测。
英文摘要:
      Objective To establish a method for determination of cholesterol in eggs by enzyme colorimetry. Methods Determination of cholesterol in eggs by enzyme colorimetry coupled with spectrophotometer at 505 nm after samples were prepared by saponification, extraction, concentration and enzyme catalyzed reaction. Results The optimal coloration temperature (37 ℃) and coloration time (15 min) had been investigated. The method had a linear range of calibration curve of 0~2.0 mg/mL for determination of cholesterol in eggs, and the linear correlation coefficients were more than 0.999. The detection limit were 1.0 mg/100 g and the relative standard deviations (RSD) were not more than 2.45%. The recoveries for the spiked samples ranged from 90.43% to 98.12%. Conclusion This method is simple and accurate, which is suitable for the determination of cholesterol in egg, duck egg, goose egg, pigeon egg and other whole egg samples or egg yolk samples.
