      目的 优化液相色谱质谱联用法测定保健食品乳清蛋白粉类中叶酸含量的前处理方法。方法 样品经过前处理, 以乙腈和0.1%甲酸溶液为流动相, 梯度洗脱, 经Agilent Eclipse XDB-phenyl柱分离, 使用液质联用仪检测。对比各个前处理方法对实验结果的影响。结果 高氯酸-碳酸钠法前处理后, 能很好地净化乳清蛋白中的蛋白质, 并对实验结果保持准确科学, 方法的精密度相对标准偏差为4.5%, 回收率为95.1%~103.6%, 相对标准偏差为3.7%。结论 采用高氯酸-碳酸钠法前处理方法能测定保健食品中乳清蛋白粉类叶酸的含量, 操作简便、准确、重现性好, 分离效果明显, 保护实验仪器和降低耗材, 确保实验结果的准确性。
英文摘要:
      Objective To optimize pretreatment methods for determination of folic acid in health food of whey protein by liquid chromatography-mass chromatography (LC-MS). Methods After being pretreated, samples were gradient eluted with acetonitrile and 0.1% formic acid as the mobile phase. Then, samples were separated by Agilent Eclipse XDB-phenyl column and detected by LC-MS. Effects of each pretreatment method on experimental results were compared. Results After being pretreated, the perchloric acid-sodium carbonate could better purify the protein in whey protein commendably, and maintained accurate and scientific experiment results. The precision of ‘of the method was 4.5%, the recovery rates were 95.1%~103.6% with a relative standard deviation of 3.7%. Conclusion The perchloric acid-sodium carbonate pretreatment can be used to determine the content of whey protein powder in health food. It is simple, accurate and reproducible operation. It also can get an obvious separation effect, protect experimental instruments, reduce consumables and ensure the accuracy of experiment results.
