      目的 建立超高效液相色谱-串联质谱法(ultra performance liquid chromatography-tandem mass spectrometry, UPLC-MS/MS)测定酒、牡蛎粉、咖啡中那非类物质含量的分析方法。方法 采用Waters HSS T3(2.1 mm×100 mm, 1.8 μm)反相色谱柱, 以0.1%甲酸水和 0.1%甲酸乙腈为流动相进行梯度洗脱, 经质谱多反应监测(multiple reaction monitor, MRM)法进行检测。结果 在10~200 ng/mL的浓度范围内, 去甲基他达拉非和硫代西地那非均表现出良好的线性关系, 2种化合物的检出限分别为50 ng/g和50 ng/mL, 定量限分别为100 ng/g和100 ng/mL。结论 该方法操作简便, 结果可靠, 灵敏度高, 可用于食品中去甲基他达拉非和硫代西地那非含量的快速测定。

      Objective To establish a method for the determination of the content of nortadalafil and thiosildenafil in wine, oyster powder and coffee by ultra performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS). Methods The samples were separated on the Waters HSST3 column (2.1 mm×100 mm, 1.8 μm) using a mobile phase of 0.1% formic acid solution and 0.1% formic acid acetonitrile with the gradient elution, and detected by multiple reaction monitor (MRM). Results In the concentration range of 10~200 ng/mL, nortadalafil and thiosildenafil both showed good linear relationships. The LODs (limits of detection) of nortadalafil and thiosildenafil were 50 ng/g and 50 ng/mL, and the LOQs (limits of quantitation) were 100 ng/g and 100 ng/mL, respectively. Conclusion This method is simple, reproducible and sensitive, which can be applied for the rapid analysis of nortadalafil and thiosildenafil in foods.
