      目的 建立一种分光光度法测定肉苁蓉多糖含量的方法。方法 肉苁蓉多糖经水提醇沉提取后, 在浓硫酸作用下水解成单糖, 并迅速脱水生成糠醛衍生物, 采用蒽酮-硫酸分光光度法在627 nm处测定其含量。结果 肉苁蓉多糖质量浓度在20~100 μg/mL范围内线性关系良好, 方法平均回收率在93.0%~99.8%之间, 相对标准偏差(relative standard deviation, RSD)为1.7%~4.5%。对肉苁蓉、管花肉苁蓉和盐生肉苁蓉3种肉苁蓉中多糖含量进行了测定, 结果表明3者的多糖含量存在差异。结论 本方法简便、准确, 可用于肉苁蓉多糖含量的测定。

      Objective To establish a method for the determination of polysaccharides in Cistanehe herba by spectrophotometry. Methods The polysaccharides were extracted with water and precipitated by alcohol. Then they were hydrolyzed into monosaccharides by sulfuric acid which was rapidly dehydrated into furaldehyde. The content of polysaccharides was determined by anthrone-sulfurine acid spectrophotometric method, and was measured at 627 nm. Results The standard curve of polysaccharide showed a good linear relationship at the range of 20~100 μg/mL. The average recoveries were 93.0%~99.8%, and the relative standard deviations were 1.7%~4.5%. There were differences in polysaccharide contents among 3 different sources of Cistanehe herba (C. deserticola, C. tubulosa and C. salsa). Conclusion This method is simple and accurate, which can be used for determination of polysaccharides in Cistanehe herba.
