目的 制备一种对豆浆中脲酶活性进行快速定性检测的试纸。方法 利用生豆浆中的脲酶能够对尿素专一性催化分解引起体系酸碱度的变化这一原理制备试纸, 能够便捷、高效、准确地对豆浆的生熟度进行判定, 从而筛查出脲酶活性为阳性的豆浆。结果 确定最优的试剂配比为10 mL 0.4 g/L苯酚红试剂和0.005 g尿素, 选择最佳的显色时间为20 min之内, 制备出的快检试纸浸入到豆浆样品后可以直接读取实验结果, 对脲酶活性进行快速判定。结论 该试纸制备方法简单, 易于操作, 检测重现性较好, 方法毒性更小, 而且在常温下能够长期储存, 可为实施豆浆的现场监督和快速检测提供有效的技术支撑。 |

Objective To prepare a rapid and qualitative test paper for determination of urease activity in soya-bean milk. Methods According to the system pH changes which was caused by selective catalytic decomposition of urease activity in soya-bean milk to urea, the prepared paper could easily, efficiently and accurately determine the maturity degree of soya-bean milk and screen the soya-bean milk with positive urease. Results The optimal ratio of reagents was 10 mL 0.4 g/L phenolred and 0.005 g urea, and the best chromogenic time was within 20 min. The prepared test paper could directly read the results after soaking into the soya-bean milk, which could quickly determine the urease activity. Conclusion The prepared paper is easy to made and operate, and has a good reproducibility, better security, and long-term storage at room temperature, which can provide a technical support for site supervision and rapid detection. |
