目的 研究食源性腹泻儿童肠道菌群的变化, 为儿童食源性腹泻的防治提供理论依据。方法 以10例食源性腹泻儿童粪便样本及10例健康儿童的粪便样本为研究对象, 提取粪便总DNA, 利用细菌16S rDNA V3区通用引物进行PCR扩增, 扩增后产物进行变性梯度凝胶电泳(denaturing gradient gel electrophoresis, DGGE)分析, 并用Quantity One软件进行分析。结果 食源性腹泻组儿童和对照组健康儿童的肠道菌群多样性存在显著性差异, 食源性腹泻组的多样性低于对照组。食源性腹泻组儿童肠道菌群具有较高的相似性并与对照组儿童肠道菌群能聚类到不同簇中。结论 食源性腹泻儿童肠道菌群多样性发生明显变化, 说明肠道菌群的多样性调控可以应用于食源性腹泻的缓解或治愈。 |

Objective To analyze the changes of intestinal flora in children with foodborne diarrhea, so as to provide the theoretical basis for the prevention and treatment of the children with foodborne diarrhea. Methods The stool samples of 10 children with foodborne diarrhea and 10 healthy children were collected as the research objects. The total DNA of stool samples were extracted, and the PCR amplification was performed using bacterial 16 s rDNA V3 area universal primers. The PCR products were analyzed by denaturing gradient gel electrophoresis (DGGE). The DGGE results were analyzed using the Quantity One software. Results There were significant differences in diversity of intestinal flora between children with foodborne diarrhea and healthy children. The diversity of foodborne diarrhea group was lower than that of control group. The intestinal flora in children with foodborne diarrhea group had a higher similarity, and was cluster into different clusters with that of healthy children. Conclusion There are obvious changes in intestinal flora diversity of children with foodborne diarrhea, which can explain the regulation of diversity of intestinal flora, and it can be applied to alleviate or cure of foodborne diarrhea. |
