目的 建立高效液相色谱紫外检测法测定配合与浓缩饲料中的叶酸含量。方法 样品经0.1 mol/L碳酸钠溶液提取, 利用EDTA对金属元素的螯合作用和甲醇沉淀蛋白的作用去除金属元素及大分子蛋白对叶酸测定的干扰, 并通过阴离子交换柱净化富集, 洗脱液浓缩定容后供高效液相色谱仪测定。结果 叶酸在0.1~250.0 μg/mL的浓度范围内具有良好的线性关系, 样品回收率为81.6%~94.6%, 相对标准偏差小于10%, 检出限可达到0.1 mg/kg, 定量限为0.3 mg/kg。结论 本方法适用于配合与浓缩饲料中叶酸含量的测定。 |

Objective To establish a method for the determination of folic acid in formula feed and concentrated feed by high performance liquid chromatography(HPLC). Methods Samples were extracted by the solution of 0.1 mol/L Na2CO3. EDTA and methanol were applied to remove the distraction of metal elements and protein molecules by reasons of chelation of EDTA to metal elements and protein deposition of methanol. Through purification by ion exchange column, the eluent was used for the determination by HPLC after concentration and constant volume. Results Folic acid had a good linear relationship in the range of 0.1~250.0 μg/mL, and the average recoveries of samples were 81.6%~94.6%. The relative standard deviation was below 10%. The limit of detection was up to 0.1 mg/kg, and the limit of quantification was 0.3 mg/kg. Conclusion This method is suitable for the determination of folic acid in formula feed and concentrated feed. |
