目的 采用高效液相色谱法检测牛奶中硫氰酸钠含量。方法 样品用5 mL乙腈沉淀蛋白质, 上清液过C18柱净化。以磷酸缓冲溶液(pH=7.0):乙腈(95:5, V:V)作为流动相, C18色谱柱(250 mm×4.6 mm, 5 μm)分析, 柱温30 ℃±2 ℃, 流速1.0 mL/min, 以紫外检测器分析样品。结果 该方法对牛奶中硫氰酸钠有较高的提取率, 质量浓度范围在5~100 mg/L内线性较好(r2=0.9995), 检测限1.0 mg/kg, 回收率>90%。结论 该方法具有预处理简单、灵敏度高的优点, 满足对牛奶中硫氰酸钠含量的检测要求。 |

Objective To detect the content of sodium thiocyanate in milk by high performance liquid chromatography (HPLC). Methods Samples were purified by protein precipitation and the C18 extraction column. Phosphate buffer: acetonitrile(95:5, V:V) (pH=7.00) was used as the mobile phase, the C18 column (4.6 mm×250 mm,5 μm) and UV detector were used to analyze samples under column temperature of 30 ℃±2 ℃ and the flow rate of 1.0 mL/min. Results The method had a high extraction rate of sodium thiocyanate in milk, the concentration of sodium thiocyanate was linear with peak area in the range of 5~100 mg/L, and the correlation coefficient was 0.9995. The limit of detection was 1.0 mg/kg and the recovery rate was above 90%. Conclusion The method is simple and sensitive, which can successfully meet the determination requirement of sodium thiocyanate content in milk. |
