目的 建立固相萃取-液相色谱串联质谱法(SPE-LC-MS/MS)同时检测食品中新红、靛蓝、喹啉黄和酸性紫(49)4种人工合成着色剂的分析方法。方法 样品经酸性水溶液提取, 混合型弱阴离子固相萃取柱净化; 采用Thermo C18色谱柱分离, 以5 mmol/L乙酸铵溶液和甲醇为流动相梯度洗脱。质谱采用电喷雾离子源多反应监测模式(MRM)分析, 外标法定量。结果 4种人工合成着色剂在0.2~1.0 mg/kg范围内呈线性相关, 相关系数均大于0.99。方法的回收率介于70.0%~101.5%之间; 相对标准偏差(RSD)(n=6)为2.3%~16.5%; 检出限均为0.20 mg/kg。结论 该方法前处理简单, 净化效果好, 具有良好的灵敏度、回收率和重现性, 适于食品中人工合成色素的检测。 |

Objective To develop a method for the simultaneous analysis of New Red, Indigo Blue, Quinoline Yellow and Acid Violet 49 in food by solid phase extraction-liquid chromatography-tandem mass spectrometry (SPE-LC-MS/MS). Methods The analytes were extracted from the samples by acidic water, then purified and enriched with a weak anion exchange SPE column. The 4 kinds of synthetic colorants were separated on a Thermo C18 column by gradient elution with 5 mmol/L ammonium acetate and methanol as the mobile phases. The analytes were determined by ESI-LC-MS/MS under multiple reaction monitoring (MRM) mode and quantified by external standard method. Results The results indicated that the calibration curve showed a good linearity of 4 kinds of synthetic colorants in the range of 0.2~1.0 mg/L (r2>0.99)。The recoveries of 4 kinds of synthetic colorants were in the range of 70.0%~101.5% with RSDs (n=6) between 2.3% and 16.5%. The detection limits were all 0.20 mg/kg. Conclusion This method is suitable for the detection of synthetic colorants in food with simple preparation, good cleaning effect, high sensitivity and better repeatability. |
