      目的 研究补充不同剂量、不同接触时间的维生素E对致敏SD大鼠机体敏感性的影响。方法 取8周龄雌性SD大鼠, 按体重随机分为5组, 每组25只, 每天经口分别给予不同剂量的维生素E(2.3、6.9、13.8 mg/kg), 在第2周、3周、4周、5周、6周时, 每组分别取出5只动物, 腹腔注射给予10 mg/只OVA致敏, 一周后使用50 mg/只OVA刺激, 检测各组动物血清中总IgE、特异性IgE和IgG1、血浆中组胺、mMCP-1和细胞因子(IFN-γ, IL-4)的水平, 大刺激后处死动物, 取心脏、肝、脾、肾、胸腺、空肠、肠系膜淋巴结等脏器, 并对这些脏器进行组织病理学观察。结果 经口给予高剂量维生素E 6周, OVA产生的免疫学反应强度受到了显著的抑制作用, 总IgE、特异性IgE、特异性IgG1、IL-4以及组胺水平显著降低, IFN-γ水平显著升高, 随接触时间的延长呈增高的趋势。且高剂量处理组动物免疫学反应强度显著低于OVA对照处理组, 对致敏SD大鼠的机体敏感性起到了有效的调节作用。结论 维生素E能够降低致敏动物机体对卵白蛋白的敏感性, 也抑制了免疫系统对卵白蛋白产生的免疫学反应。

      Objective To study the influence of vitamin E on the susceptibility in sensitized SD rats. Methods SD rats, 8-week-old, were randomly divided into 5 groups (25 in each group). Rats were oral administrated of different doses of vitamin E (2.3, 6.9, 13.8 mg/kg). To sensitize the rats, 5 in each group were given 10 mg/rat of OVA intraperitoneally at the end of week 2, 3, 4, 5 and 6, respectively. One week later, rats were challenged with 50 mg/rat. Then the levels of total IgE, OVA-specific IgE and IgG1, histamine, mMCP-1 and cytokines in serum of each group were examined. Rats were sacrificed and their heart, liver, spleen, kidney, thymus, jejunum and mesenteric lymph nodes were collected for pathologic analysis. Results The immunological response induced by OVA was inhibited after 6-week oral administration of vitamin E. The levels of total IgE, OVA-specific IgE and IgG1, histamine and IL-4 were decreased while IFN-γ was increased. In addition, the reaction of rats in the group of high dose were weaker than the group of low dose. All these results suggested vitamin E could regulate the sensitivity of SD rat effectively. Conclusion Vitamin E can inhibit anaphylactic reaction induced by antigen in sensitized rats in a dose-dependent manner.
