目的 对采用ELISA方法测定花生油中黄曲霉毒素B1含量的不确定度进行分析, 找出影响不确定度的因素, 为评价其测定方法和检测结果的可靠性提供依据。方法 根据JJF 1059.1-2012《测量不确定度评定与表示》的规定, 对测定过程中标准曲线拟合、重复测量、样品的稀释、样品的称量、回收率等引入的不确定度分量进行评定。结果 测定结果合成不确定度为0.45 μg/kg, 扩展不确定度为0.90 μg/kg, 花生油中黄曲霉毒素B1含量测定结果为(16.54±0.90) μg/kg (k=2)。结论 测定结果不确定度主要由标准曲线拟合所引入, 所建立的不确定度评定方法可用于ELISA方法测定花生油中黄曲霉毒素B1含量结果的判断。 |

Objective To analyze the uncertainty of aflatoxin B1 content determined by enzyme-linked immunoassay (ELISA) and find out the influential factors of uncertainty to provide the reliable basis for this method and detection result. Methods Based on JJF 1059.1-2012 “Evaluation and expression of uncertainty in measurement”, the uncertainty components that introduced from repetition measurements, working charac-teristic fitting, electron balance and recovery rate were evaluated in the process of determination. Results The results showed that the combined uncertainty of aflatoxin B1 was 0.45 μg/kg, the expanded uncertainty was 0.90 μg/kg, and the content of aflatoxin B1 in paenut oil was (16.54±0.90) μg/kg (k=2). Conclusion The uncertainty is mainly introduced by standard curve fitting, and this uncertainty evaluation method can be used for judging the result of the determination of aflatoxin B1 in peanut oil by ELISA. |
