      目的 研究茶多酚等天然抗氧化剂对香烟烟雾致损的HEK293细胞的保护效能及JAK2/STAT3 代谢调控机制。方法 香烟过滤嘴中添加茶多酚、白芦藜醇、维生素C、芦丁、槲皮素、绿原酸、花青素、原花青素8种植物提取物后收集烟气, 对 HEK293 细胞进行 MTT 对比研究及运用 Western blot 技术分析其Bcl-2、Bax、STAT3蛋白表达量变化。结果 香烟烟雾可降低HEK293细胞存活率, 呈浓度依赖关系; 上调 HEK293 细胞中 Bcl-2 蛋白相对表达量, 下调 Bax、STAT3 蛋白相对表达量, 且各蛋白变化趋势呈浓度依赖效应; 在过滤嘴中添加茶多酚、白芦藜醇、维生素C、芦丁、槲皮素、绿原酸、花青素、原花青素8种抗氧化剂明显抑制了香烟烟雾导致HEK293细胞死亡, 结果具有显著性差异(P<0.05), 其中以茶多酚效果最佳。结论 8种植物提取物均能对香烟所致HEK293细胞损伤产生一定的保护作用。

      Objective To investigate the metabolic regulation mechanism of tea polyphenols and other natural antioxidants antagonizing cigarette smoke damaging HEK293 cells and its JAK2/STAT3. Methods Eight kinds of antioxidants, tea polyphenols, resveratrol, vitamin C, rutin, quercetin, chlorogenicacid, antho-cyanins and proanthocyanidins, were added to cigarette filters and the smoke was collected. HEK293 cells were studied by MTT trail data and the protein expression Bcl-2, Bax, and STAT3 were analyzed by Western blot technique. Results Cigarette smoke could reduce the HEK293 cell survival rates in a dose-dependent manner. With up-regulate Bcl-2 protein relative expression, down-regulate Bax, and STAT3 protein relative expression levels, each protein expression levels were in a dose-dependent effect for HEK293 cells. Comparing to the control, adding 8 kinds of antioxidants to cigarette filters could inhibit the increase of HEK293 cells death rates caused by cigarette smoke obviously. The results had significant differences (P<0.05), and tea polyphenols was the best. Conclusion Eight kinds of antioxidants can protect HEK293 cell from damage caused by cigarettes smoke.
