目的 利用纳米金在盐诱导下凝聚后引起的颜色变化,建立基于非标记的AFB1特异性结合适配体,针对AFB1的灵敏快速可视化检测方法。方法 通过对适配体DNA浓度、NaCl盐溶液、pH值等条件的优化,确定最佳反应体系并进行特异性实验。结果 本方法具有良好的灵敏度和特异性,线性范围和检测限分别是0.025—10 ng/mL和0.025 ng/mL,可以满足中国和欧盟的最大残留限量(MRL)的检测要求。结论 该方法实现了对黄曲霉毒素B1的可视化检测,简单、高效、可行,具有较高的分析灵敏度和较好的特异性。 |

Objective The colorimetric detection of AFB1 based on a label-free the AFB1 aptasensor using unmodified gold nanoparticles(AuNPs) was developed in the present study. Methods Concentration of DNA and NaCl was optimized , as well as the pH values , to determine the optimal reaction system. Specific experiment was carried out with the interference of OTA. Results The linear dynamic range and detection sensitivity were found to be 0.025-10 ng/mL and 0.025 ng/mL of AFB1 respectively, which were lower than the maximum residue limit (MRL) in edible food, as set by China and the European Commission. Conclusion This study provides a simple, sensitive and visible detection method for AFB1 analysis, which can be applied in future on-site detection for food and agricultural products. |
